Мутації в генах BRCA1/2 підвищують ризик захворювань на рак молочної залози, рак підшлункової та передміхурових залоз у чоловіків. В Україні мутації генів BRCA1/2 вивчалися лише у жінок, а ситуація з захворюваннями, пов’язаними з порушенням роботи цих генів у чоловіків залишається майже не дослідженою. Аналіз 15 чоловіків, хворих на рак передміхурової залози та 15 потенційно здорових чоловіків того ж віку на наявність двох мутацій гену BRCA1 (185delAG, 5382insC) та однієї мутації в гені BRCA2 (6174delT) проводили методом мультипраймерної ПЛР. Жоден з проаналізованих пацієнтів не характеризувався наявністю вказаних мутацій, що відповідає даним літератури, які свідчать про низьку частоту цієї події у чоловіків Північної Америки, Австралії, Азії, а також Західної, Центральної та Південної Європи.

Мутации в генах BRCA1/2 повышают риск возникновения рака молочной железы, рака поджелудочной и предстательной желез у мужчин. В Украине мутации генов BRCA1/2 изучались только у женщин, а ситуация с данными заболеваниями, связанными с нарушением работы этих генов у мужчин остается почти не исследовано. Анализ 15 мужчин, больных раком предстательной железы и 15 потенциально здоровых мужчин того же возраста на наличие двух мутаций гена BRCA1 (185delAG , 5382insC) и одной мутации в гене BRCA2 (6174delT) проводили методом мультипраймерной ПЦР. Ни один из проанализированных пациентов не характеризовался наличием указанных мутаций, что соответствует данным литературы, которые свидетельствуют о низкой частоту этого события у мужчин Северной Америки, Австралии, Азии, а также Западной, Центральной и Южной Европы.

Mutations in BRCA1/2 genes are known to increase the risk of human breast cancer, pancreatic cancer and prostate cancer. In Ukraine only mutations in woman BRCA1/2 were studied, thus the situation with these diseases related to disturbance of these genes in ukrainian men remains poorly explored. We performed PCR analysis on 15 men with prostate cancer and 15 potentially healthy men of the same age in order to detect presence of two mutations in BRCA1 gene (185delAG, 5382insC) and one mutation in BRCA2 gene (6174delT). None of the analyzed patients demonstrated the presence of these mutations, which accords to the literature data that specify a low frequency of this event in men in North America, Australia, Asia, and the Western, Central and Southern Europe.

The BRCA1/2 genes play important role in the control of cell cycle, DNA repair, regulation of transcription and apoptosis. A disorder in these genes leads to various pathologies in humans, particularly breast cancer (BC) in women, which is one of the most prevalent malignancies and the main cause of death from cancer  in women around the world. About 10% of patients with breast cancer are characterized by the presence of mutations in BRCA1  and BRCA2 (acronyms stand for “BReast - CAncer susceptibility gene” 1 and 2 respectively).

The probability of breast cancer occurrence given presence of these mutations is estimated, according to different authors, at 50 to 90% the literature data also indicate that mutations in these genes are associated with the increased risk of breast cancer, pancreatic cancer and prostate cancer in men; the course of disease in male-carriers of BRCA1/2 mutations is more complicated though.

In Ukraine only mutations in women's' BRCA1/2 were studied, thus the situation  related to disturbance of these genes in Ukrainian men population remains poorly explored. Thereby, the aim of the study was to come up with methods and preliminary analyses of the mutations’ frequency in BRCA1/2 genes in men with  prostate cancer (PC) (as  the disease for which  its’  association with  mutations in BRCA1/2 genes is shown), and the control group of potentially healthy men. According to the literature, the prevalence of prostate cancer in the world is estimated at 2.4%, whereas the BRCA1 mutations make up to 2.3% patients with prostate cancer and 1.9% for control individuals; for BRCA2 mutation – 3.3% for patients with the disease and 1.6% for controls.

The study included 15 men with prostate cancer and 15 potentially healthy men of age ranging from 59 to 75 years. DNA extraction was performed using a phenol-chloroform method. In order to detect presence of two mutations in BRCA1 gene (185delAG, 5382insC) and one mutation in BRCA2 gene (6174delT) we performed PCR analysis using respective specific primers. For each mutation we used three pairs of primers (one common, one specific to the mutation and specific to the wild-type allele) according to the literature data and by self-optimization. Following amplification 4 μl of the PCR product was applied to 8% polyacrylamide gel and electrophoresis procedure was performed to assess the presence and length of the product.

The results of DNA analysis of 15 patients with prostate cancer and 15 potentially healthy men indicated that none of the investigated individuals  demonstrated the presence of these mutations.

Since the mutations in BRCA1/2 genes are associated not only  with prostate cancer in men, but also with other common forms of cancer, so that accords to the literature data that specify a low frequency of this event in men in the world. Hence the further continuation of our study with bigger sample size will not only find out the frequency of such events in Ukraine in general, but also (by determining the specific type of mutation) will predict the prospects of treating the individual patient in particular.
