Involvement of the liver in protein metabolism is characterized by active processes of synthesis and breakdown of proteins that are essential for the body. In the liver about 13-18 grams of proteins are synthesized per day. Of these, albumin, fibrinogen, prothrombin are produced only in the liver. In addition, it is synthesized to 90 % alpha-globulin and 50 % gamma-globulin of an organism. Liver cells have a complete set of enzymes necessary for the transformation of amino acids and their degradation, modification and synthesis of new nitrogen compounds. "Nonessential" amino acids and nitrogenous bases of nucleic acids, which promote the formation of protein reserve of the organism under the condition of the limited receipt are formed from simple precursors in the liver [1-3, 7, 8]. Thus, derangement of protein metabolism may lead not only to loss of cell activity, but the whole organism. To prevent it, a timely receipt is needed to the organism of structural components for the synthesis of new proteins. With the purpose of proceeding in functioning of protein metabolism a timely receipt is needed to the organism from outside structural components – amino acid and other biologically active substances – correctors of protein metabolism [8]. As a source of these substances can be used tablets «Medikabol», based on an extract of Medicago sativa sowing grass [5, 6], which explains the feasibility of studying its impact on functional status of hepatocytes in rats under acute liver injury induced by CCL4 [2, 3]. Purpose – to examine the influence of drugs and «Medikabol» comparison potassium orotate and Silibor the state of protein metabolism in acute hepatitis tetrahlormetanovoho in rats. Materials and methods The effects of tablets «Medikabol» to functional state of protein metabolism was studied in 40 white mongrel rats weighing 180-200 g under acute liver injury [4 ]. The experimental animals were randomized into 5 groups: 1 – Intact control (IC ), 2 – control pathology (CP), 3 – Experimental receiving «Medikabol» in a dose of 25 mg / kg, 4 – Experimental receiving comparator – Potassium orotate in doses of 180 mg / kg, 5 – Experimental receiving comparator Silibor at a dose of 25 mg / kg. The functional status of hepatocytes and protein metabolism in rats was assessed by the percentage of survival of experimental animals, the mass ratio of the liver, biochemical parameters in serum [4]: the level of total protein, the average molecular weight, albumin, urea, values of oxidative modification of proteins [4], the activity of cytolytic enzymes alanine aminotransferase, aspartate aminotransferase [1], g - glutamiltransferase [1]. Intensity of lipid peroxidation and antioxidant defense status was evaluated by the content of TBA- active products, catalase and glutathione (G-SH) in liver homogenate [4 ]. The obtained results and their discussion. Experimental data suggest the development of metabolic and structural disturbances caused by the introduction of hepatotoxine carbon tetrachloride. Introduction to experimental animals tablets «Medikabol» in a dose of 25 mg / kg and reference preparations of potassium orotate in doses of 180 mg / kg and Silibor at a dose of 25 mg / kg in a health care background mode on toxic liver injury has led to positive changes, as evidenced by corresponding changes of the main functional and biochemical parameters. Thus, the use of the investigational product and the comparator helped reduce the severity of necrotic and degenerative changes in the parenchyma of the liver that is caused by inhibition of lipid destructive processes. Inhibition of lipid peroxidation processes in the application of tablets «Medikabol» was reflected in the trend compared with the CP group to restore the functional capacity of the AOS and AOP – GSH level and catalase activity in liver homogenate are increased to 19,18 and 20,60 %, and 9,73 and 6, 85%, respectively. The use of tablets «Medikabol» in a dose of 25 mg / kg in health care regime unlike referent drugs potassium orotate in doses of 180 mg / kg and Silibor in a dose of 25 mg / kg are facilitated the resumption of protein metabolism and warned degradation of membrane and cellular proteins in terms acute CCl4 hepatitis. Conclusions. 1. It was established that an acute toxic liver damage in rats induced by CCl4, leads to disruption of structural and functional activity of hepatocytes. 2. The use of tablets «Medikabol» in a dose of 25 mg / kg based on an extract of Medicago sativa sowing grass crop health care regime restoring processes of protein metabolism and prevents degradation of protein molecules through the contents of large amounts of protein, amino acids and other biologically active substances, which determine its antioxidant, membrane-stabilizing, organ protective and anti-inflammatory properties. 3. It was found that the level of pharmacological activity of studied herbal product almost in 2 times was greater than the referent drug potassium orotate in doses of 180 mg / kg. Література
